As far as I know, DLC is tied to the account (CD keys are input). If you use Steam, there's a different method used, and they'll redownload.
You can just copy them out to another folder, and you can just drag and drop the files from that backup to the new install.
If you're worried, make a copy of: %APPDATA%\Tropico 4
This will save your profile, saves, and, for non-Steam versions, DLC files (which should redownload anyway).
Just copy it back there when you're done (Start -> run %APPDATA%, make a new folder "Tropico 4").
For Steam versions, you can back-up the Tropico 4 DLC bits and pieces:
For x64: C:\Program Files (x86)\Steam\steamapps\common\tropico 4\dlc
For x86: C:\Program Files\Steam\steamapps\common\tropico 4\dlc
For mods if you followed my install instructions, you'll find them in:
For x64: C:\Program Files (x86)\Steam\steamapps\common\tropico 4\packs\boot\persist
For x86: C:\Program Files\Steam\steamapps\common\tropico 4\packs\boot\persist
Inside there, you'll find "Game.hpk" and "Shaders.hpk" which are part of the game -- the rest are mods.
